Antagonistic interactions between benzo[a]pyrene and C60 in toxicological response of Marine Mussels ArchiMer
Barranger, Audrey; Langan, Laura M; Sharma, Vikram; Rance, Graham A; Aminot, Yann; Weston, Nicola J; Akcha, Farida; Moore, Michael N; Arlt, Volker M; Kholbystov, Andrei N; Readman, James W; Jha, Awadhesh N..
This study aimed to assess the ecotoxicological effects of the interaction of fullerene (C60) and benzo[a]pyrene (B[a]P) on the marine mussel, Mytilus galloprovincialis. The uptake of nC60, B[a]P and mixtures of nC60 and B[a]P into tissues was confirmed by GC-MS, LC-HRMS and ICP-MS. Biomarkers of DNA damage as well as proteomics analysis were applied to unravel the toxic effect of B[a]P and C60. Antagonistic responses were observed at the genotoxic and proteomic level. Differentially expressed proteins (DEPs) were only identified in the B[a]P single exposure and the B[a]P mixture exposure groups containing 1 mg/L of C60, the majority of which were down-regulated (~52%). No DEPs were identified at any of the concentrations of nC60 (p < 0.05, 1% FDR)....

Biomonitoring of Pb and Cd in two impacted watersheds in Southeast Brazil, using the freshwater mussel Anodontites trapesialis (Lamarck, 1819) (Bivalvia : Mycetopodidae) as a biological monitor BABT
Tomazelli,Andréa C.; Martinelli,Luiz A.; Avelar,Wagner E. P.; Camargo,Plínio B. de; Fostier,Anne-Helene; Ferraz,Epaminondas S. B.; Krug,Francisco J.; Santos Júnior,Dário.
Studies were carried out to investigate the contamination of Piracicaba and Mogi-Guaçu River basins (São Paulo State, Southeastern, Brazil) for heavy metals (Cd and Pb), using the limnic bivalve Anodontites trapesialis as a biological monitor. The results showed that the concentrations of Pb were higher than the control group in both basins, showing the bioavailability of this non-essential element in the basins. The concentrations were higher in the Mogi-Guaçu than in the Piracicaba basin, and in the slightly contaminated sites in both basins. There was no correlation between the degree of human impact and Cd and Pb concentrations, it was not possible to infer about concentrations of these heavy metals in the bivalves based only in a broad evaluation of...

Characterization of ten microsatellite loci in the blue mussel Mytilus edulis ArchiMer
Lallias, Delphine; Stockdale, Ruth; Boudry, Pierre; Lapegue, Sylvie; Beaumont, Andy R..
Mussels of the genus Mytilus are ecologically and commercially important worldwide, and they form hybrid complexes where their distributions overlap. Allozyme and nuclear markers have bee It Used to investigate their genetics over many years, but successful development of reliable highly valuable microsatellite markers has lagged be hind other shellfish species. We have developed and characterized tell novel microsatellite loci that amplify reliably for the blue mussel Mytilus edulis. The number of alleles among 30 individuals from a wild population (Menai Strait, North Wales UK) ranged between 9 and 29 and the observed heterozygosity between 0.300 and 0.954. Significant heterozygote deficiencies against the Hardy-Weinberg model were observed at 6 out of...

Chemical environment of the hydrothermal mussel communities in the Lucky Strike and Menez Gwen vent fields, Mid Atlantic ridge ArchiMer
Sarradin, Pierre-marie; Caprais, Jean-claude; Riso, Ricardo; Kerouel, Roger; Aminot, Alain.
Water samples were collected around the communities of a hydrothermal mussel, Bathymodiolus sp., in the Lucky Strike (1700 m depth) and Menez Gwen (850 m depth) areas, Mid Atlantic Ridge, and analysed for chemical constituents. The environment surrounding the organisms consists in sea water (88 to 100%) mixed with hydrothermal fluid with pH between 6.2 and 8, Sigma S (H2S + HS-) concentrations from 0 to 62 mu mol l(-1). High dissolved organic carbon (DOC) concentrations suggest a highly productive ecosystem. Production of ammonium and DOG, consumption of nitrate and sulfide in the vicinity of the organisms are among the environmental changes found in relation with local biological activity. Concentrations of Cu and Pb were high, implying that the organisms...

Comment améliorer la précocité de l'alerte en conchyliculture ? Exemple des mortalités de moules en 2014 dans les Pertuis Charentais ArchiMer
Lupo, Coralie; Prou, Jean.
Surveillance of shellf ish health is based on clinical surveillance of shellfish mortality, including a detection phase and a mandatory notification phase. While control measures are limited, early detection of pathogens is a challenge which needs to be met. The relevance and feasibility of an enhanced clinical surveillance based on phone interviews were studied. The results made it possible to reconstruct the spatial and temporal evolution of the outbreaks of mussel mortality which occurred in Pertuis Charentais in 2014. Moreover, a qualitative analysis of the interviews revealed contextual elements that were used to enhance the previous description and to devise causal scenarios which could be used as research hypotheses. By incorporating the...

Contamination de moules (Mytilus sp.) en milieu marin par des substances pharmaceutiques et produits de soin ArchiMer
Bachelot, Morgane.
Pharmaceutical and personal care products used for human health protection are detected in all aquatic ecosystems. Among them, some compounds have physico-chemical or pharmacological caracteristics that suggest a bioaccumulation capability. It’s the case of the UV filters, EHMC and OC, with log Kow higher than 5 or pharmaceuticals like diazepam and tetrazepam, with log Kow higher than 2.82. The aim of these Phd works was to assess marine mussels contamination for UV filters and benzodiazepines. So, a new analytical method was developed and validated for the determination of these compounds in mussels. EHMC and OC analysis realized in mussels sampled in french coastal waters showed that mainly contamination came from recreational activities in summer...

Détermination de Valeurs Guides Environnementales (VGE) mollusques alternatives aux Normes de Qualité Environnementale (NQE) définies dans la DCE ArchiMer
Sire, Alizee; Amouroux, Isabelle.
The Water Framework Directive (DCE 2000/60/CE), aims to achieve the good conditions (chemical and ecological) of different waters in Europe. The Environmental Quality Standards (EQS) are the tools used to evaluate the chemical condition waters. Initially, the EQS are defined for the water matrix, but with the analytical difficulties and low spatial and temporal representativity of water samples, the monitoring of contaminants must be carried out on alternative matrices, as mollusks and particularly the mussels. Among the 45 priority and dangerous priority substances establishes by the WFD 2013/39/UE, 25 substances are hydrophobic and bioaccumulable. The main goal of this study is to determine thresholds applicable in the mussels at least as protective as...

Kamel, Naouel; Burgeot, Thierry; Banni, Mohamed; Chalghaf, Mohamed; Devin, Simon; Minier, Christophe; Boussetta, Hamadi.
This study examined the influence of increasing temperatures in spring and summer on biochemical biomarkers in Mytilus galloprovincialis mussels sampled from Bizerte lagoon (northern Tunisia). Spatial and seasonal variations in a battery of seven biomarkers were analyzed in relation to environmental parameters (temperature, salinity and pH), physiological status (condition and gonad indexes), stress on stress (SoS) and chemical contaminant levels (heavy metals, PAHs and PCBs) in digestive glands. Integrated Biological Response (IBR) was calculated using seven biomarkers (AChE, BPH, MXR, GST, CAT, MDA and MT). Seasonal variations in biological response were determined during a critical period between spring and summer at two sites, where chemical...

Mass mortality outbreaks (90-100%) in both juvenile and adult blue mussels were reported in 2014 along the Pertuis Charentais area of the French Atlantic coast. A series of studies were carried out to learn more about the phenomenon through detecting pathogens, describing environmental conditions and establishing the physiological status of the animals. Bacteria identif ied as belonging to the V. splendidus species were detected in dying mussels and seem able to cause mortality in blue mussels in the laboratory. Unusual environmental conditions (large amounts of fresh water, re-suspension of sediments during a series of storms, and a high renewal rate of specific water masses in Brittany’s Pertuis area) may be partially responsible for sparking the phenomen

Estimation du stock de moules sur bouchots en baie du Mont Saint-Michel ArchiMer
Gerard, Denis.
The quantity of mussels cultivated on wooden poles called « bouchots », in Mont Saint-Michel bay, has been estimated in april 2002, by volume, number and weight. To do so, a sampling strategy has been conceived, operated and then optimized, taking into account the spatial distribution of this stock and the material and human constraints (especially the amount of persons and working days available). The sampling plan uses procedures of stratification, degrees, regression, so as to reach the optimum precision related to cost. The operation took a great advantage of the efficient help of mussel farmers. During the first day, the task consisted to estimate the number and position of poles occupied by mussels, with the help of 5 persons on boat. The following 2...

Farming systems as an ecological refuge against the parasitism by Edotia doellojuradoi (Isopoda: Idoteidae) on Mytilus edulis (Mollusca: Mytilidae) in Aysén, Chile RChHN
VALENCIA,VALENTINA; GEORGE-NASCIMENTO,MARIO.
This study compares the prevalence and abundance of the isopod Edotia doellojuradoi in the mussel Mytilus edulis, between water column rope farming systems and nearby natural benthic beds at Puerto Raúl Marín Balmaceda (43° 46' S, 72° 57' W), Aysén, Chile, taking into account the maximum shell length of the host as a covariate. In addition, the damage caused by E. doellojuradoi was assessed with the visual inspection of the gills, according to the protocol of Zaixso et al. (2009). The isopod was found almost exclusively in natural benthic habitats, where the prevalence increased with the length of the host. The isopod is a macroparasite and not a commensal as previously suggested because it causes gill damage to the mussel, which increased with the...

Genome mapping in shellfish aquaculture species ArchiMer
Lapegue, Sylvie.
The development of fish and shellfish farming is facing several challenges linked to the infancy of the industry: i.e. domestication, genetic improvement of disease resistance, genetic improvement of feed efficiency to fish meal and fish oil substitution by vegetal products, development of adapted vaccines, supply of fry or spat at high quality and high robustness, adaptation to environmental change, production of healthy, high quality consumer products and limitation of environmental impact of the activity and escapees. Over recent years, the EU and individual countries have supported the development of genomic tools for some aquaculture species (e.g. rainbow trout, sea bass, sea bream, turbot pacific oyster, Atlantic salmon). New perspectives arise...

Genomic abnormalities affecting mussels (Mytilus edulis-galloprovincialis) in France are related to ongoing neoplastic processes, evidenced by dual flow cytometry and cell monolayer analyses ArchiMer
Benabdelmouna, Abdellah; Saunier, Alice; Ledu, Christophe; Travers, Marie-agnes; Morga, Benjamin.
In the context of the abnormal mass mortality of mussels in France since 2014, Flow CytoMetry (FCM) was used in 2015 and 2016 to study the DNA content and cell cycle characteristics of hemic circulating cells collected from 2000 mussels. The mussels were sampled from 12 wild and cultivated blue mussels stocks distributed along the French Atlantic coast from the south Brittany to Pertuis Charentais areas. During these surveys, various genetic abnormalities were frequently detected, and ploidy characteristics revealed contrasting profiles that corresponded to respective contrasting sanitary status, i.e. healthy mussels with high cytogenetic quality (HCQ) versus diseased mussels with low cytogenetic quality (LCQ). In the present work, FCM and hemocytology...
